Lots of people are courageous

Lots of people are courageous enough to tell their managers what (often unrecognized) risk is attached to decisions that are being made. It is also fairly common that the decision maker is completely unqualified to recognize this risk but has every reason to trust and respect the judgement of the person who is advising them that the risk/benefit is unfavorable. However, very often that professional integrity does NOT work out well for the cosncientious advisor as the possible negative consequences will be unpopular with the bosses' bosses and those guys aren't going to take the heat for that. So the conscientious advisor is perceived and portrayed in a negative light, the decison that conforms with the illusions of the top bosses IS made and enforced, and when the invevitable failure eventually does occur, the poor saps who were trying to do the right thing are even blamed as the cause of the failure. They end up fired and the boss puts in for his bonus. Yay.

Please spare me all your accusations of "paranoid ranting" and "internet diva-nation" all you stooges out there. This is exactly what happens all the time in complex corporate environments, especially ones with multi-layer executive hierarchies wherein entree into the executive suite is actually and exclusively a political decision however much it is purported to be a function of professional accumen (Life Tech, e.g. but very very common throughout the US).

And not to minimize the tragedy of many lives destroyed every day by these kind of machinations in corporate America, for an especially poignant example, go read up on the history of the Challenger disaster and then read my first paragraph again.

Even if they get to accurate sub-femtoliter reaction volumes, single proton detection is still "seeing" a +1 proton increase in a pool of a few dozen existing protons at pH 7. At those levels, whether or not there are 1, 2 or 3 polymerase molecules in the reaction will (based on the pI of the polymerase) greatly alter the buffering capacity.

It's very suspect because there is a milestone payment due. Yet, if milestones are not reached, PR falls short of all their crazy claims 7 months ahead of reality. So everything that is said must be suspect and no one really knows how milestones are reached. Shareholders really need to look at this one. It's a whopper.

The last interview I saw with

The last interview I saw with product manager this week is that they are routinely getting 400bp now, they promised 200bp by year end and they delivered 100 bp if that.

So what is it ? The R & D lab keeps doing better but the customers can't do crap. Start from the start please... Long reads are really going to be a killer app, stop talking about chip size, molecular diag labs don't give a crap about exomes or RNA seq.

No, machines placed in Dec '10 and Jan '11. This was the data from commercial machine, not beta. PGM has some development time to be useful. Even if that run was only $500, it still took days to set up. The enrichment step also needs a lot of work.

That is beyond awful. 4.7 Mb of Q20 data for $1000? My last run on an Illumina machine was about $8 for that amount of data at Q35. With a longer mean read length. And close to 95% mappable.

There is no way LIFE can charge for machines and reagents with that quality of output.
